CHMP2A is a core component of the endosomal sorting required for transport complex III (ESCRT-III), which is involved in multivesicular bodies (MVBs) formation and sorting of endosomal cargo proteins into MVBs. Removing 42 amino acids from the C-terminal of the protein unmasks a common ability to associate with endosomal membranes and assemble into large polymeric complexes, and the C-terminal sequences are also important for interaction of ESCRT-III protein with VPS4. Thus, ESCRT-III proteins cycle between a closed state and an activated open state is under control of sequences at the C-terminus and associated factors.
