Descriptions

FLT3 is a tyrosine-protein kinase that acts as cell-surface receptor for the cytokine FLT3LG and regulates differentiation, proliferation and survival of hematopoietic progenitor cells and of dendritic cells. The juxtamembrane (JM) region (between the transmembrane helix and the cytoplasmic kinase domain) negatively regulates catalytic activity in various receptor tyrosine kinases, and this autoinhibition is relieved through ligand-stimulated Tyr phosphorylation. Internal tandem duplication (ITD) mutations within the JM domain contribute to about 17%-34% of FLT3-activating mutations in acute myelogenous leukemia (AML). These repeat sequences serve to disrupt the autoinhibitory activity of the JM domain, resulting in the constitutive activation of FLT3. Also, point mutations such as Asp835 or Asn841 within the activation loop of the FLT3 kinase domain lead to constitutive activation of the receptor, resulting in FLT3 autophosphorylation and phosphorylation of downstream targets.

3 GO annotations of cellular component

Name Definition
CSF1-CSF1R complex A protein complex consisting of a macrophage colony-stimulating factor (CSF1, also called M-CSF) dimer bound to a dimerized receptor (CSF1R, also called FMS). Receptor dimerization requires the presence of the ligand.
plasma membrane The membrane surrounding a cell that separates the cell from its external environment. It consists of a phospholipid bilayer and associated proteins.
receptor complex Any protein complex that undergoes combination with a hormone, neurotransmitter, drug or intracellular messenger to initiate a change in cell function.

4 GO annotations of molecular function

Name Definition
ATP binding Binding to ATP, adenosine 5'-triphosphate, a universally important coenzyme and enzyme regulator.
cytokine binding Binding to a cytokine, any of a group of proteins that function to control the survival, growth and differentiation of tissues and cells, and which have autocrine and paracrine activity.
growth factor binding Binding to a growth factor, proteins or polypeptides that stimulate a cell or organism to grow or proliferate.
macrophage colony-stimulating factor receptor activity Combining with macrophage colony-stimulating factor (M-CSF) receptor ligand and transmitting the signal from one side of the membrane to the other to initiate a change in cell activity by catalysis of the reaction

28 GO annotations of biological process

Name Definition
developmental pigmentation The developmental process that results in the deposition of coloring matter in an organism, tissue or cell.
immune response in brain or nervous system An immune response taking place in the brain or nervous system.
inflammatory response The immediate defensive reaction (by vertebrate tissue) to infection or injury caused by chemical or physical agents. The process is characterized by local vasodilation, extravasation of plasma into intercellular spaces and accumulation of white blood cells and macrophages.
innate immune response Innate immune responses are defense responses mediated by germline encoded components that directly recognize components of potential pathogens.
lipid metabolic process The chemical reactions and pathways involving lipids, compounds soluble in an organic solvent but not, or sparingly, in an aqueous solvent. Includes fatty acids; neutral fats, other fatty-acid esters, and soaps; long-chain (fatty) alcohols and waxes; sphingoids and other long-chain bases; glycolipids, phospholipids and sphingolipids; and carotenes, polyprenols, sterols, terpenes and other isoprenoids.
macrophage chemotaxis The movement of a macrophage in response to an external stimulus.
melanocyte differentiation The process in which a relatively unspecialized cell acquires specialized features of a melanocyte.
melanocyte migration The orderly movement of melanocytes from one site to another, often during the development of a multicellular organism. A melanocyte is a pigment cell derived from the neural crest. It contains melanin-filled pigment granules, which give a brown to black appearance.
microglia development The process aimed at the progression of a microglial cell over time, from initial commitment of the cell to a specific fate, to the fully functional differentiated cell.
microglia differentiation The process in which a relatively unspecialized cell acquires specialized features of a microglial cell. Microglia are glial cells that act as the immune cells of the central nervous system. They form part of the supporting structure of this system.
osteoclast development The process whose specific outcome is the progression of a osteoclast from its formation to the mature structure. Cell development does not include the steps involved in committing a cell to a specific fate. An osteoclast is a specialized phagocytic cell associated with the absorption and removal of the mineralized matrix of bone tissue.
osteoclast differentiation The process in which a relatively unspecialized monocyte acquires the specialized features of an osteoclast. An osteoclast is a specialized phagocytic cell associated with the absorption and removal of the mineralized matrix of bone tissue.
osteoclast maturation A developmental process, independent of morphogenetic (shape) change, that is required for an osteoclast cell to attain its fully functional state. An osteoclast is a specialized phagocytic cell associated with the absorption and removal of the mineralized matrix of bone tissue, and which typically differentiates from monocytes.
