Descriptions

The family of death-associated protein kinases (DAPKs) comprises five enzymes that exhibit a high degree of sequence homology in their catalytic domains. These enzymes function in the triggering of cellular apoptosis. DAPK2 is a CaM-dependent protein kinase (CaMK) that is kept in an inactive state by a double-locking mechanism. This mechanism requires dephosphorylation of an autophosphorylated residue within the CaM-binding domain, and is disrupted by CaM in response to Ca2+ stimuli. The autoinhibitory domain is a structural feature that regulates DAPK2, a pro-apoptotic serine/threonine kinase that controls the kinase’s activity through autoinhibition. This autoinhibition is released by CaM binding to its CaM-binding domain.
