Caseinolytic metallopeptidase (Selecase) is a protein that was currently characterized as a novel protein, which was extremely soluble and shows reversible and concentration-dependent equilibrium between different discrete states. Selecase exhibits reversible autoinhibition through concentration-dependent oligomerization. This autoinhibition is due to sequestering the enzyme in oligomeric forms (dimers, tetramers, octamers) that are catalytically incompetent. The transitions between these forms involve major rearrangements, particularly affecting the C-terminal subdomain, and are reversible by simple dilution. At the concentration between 0.2 and 0.3 mg/ml, monomeric selecase becomes an active state that oligomers correspond to self-inhibiting species. In contrast, higher enzyme concentrations yield lower activity.
