Descriptions

IRF3 is a key transcriptional regulator of type I interferon (IFN)-dependent immune responses and is present in an inactive cytoplasmic form. The C-terminal domain of IRF-3 residues participates in the autoinhibition of IRF-3 activity via an intramolecular association with the N-terminal region. Activation of IRF3 depends on the kinase binding sites within the C-terminal region of the autoinhibitory region. The IRF3 N-terminal and C-terminal segments interact with each other to conceal the H3 and H4 helices in the IFN association domain; the hydrophobic residues in H3 and H4 are involved in DNA binding following AD translocation after IRF3 activation. The C-terminal autoinhibitory region contains phosphorylation sites and the phosphorylation of the sites converts IRF3 into a constitutively active form.
