FLT3 is a tyrosine-protein kinase that acts as cell-surface receptor for the cytokine FLT3LG and regulates differentiation, proliferation and survival of hematopoietic progenitor cells and of dendritic cells. The juxtamembrane (JM) region (between the transmembrane helix and the cytoplasmic kinase domain) negatively regulates catalytic activity in various receptor tyrosine kinases, and this autoinhibition is relieved through ligand-stimulated Tyr phosphorylation. Internal tandem duplication (ITD) mutations within the JM domain contribute to about 17%-34% of FLT3-activating mutations in acute myelogenous leukemia (AML). These repeat sequences serve to disrupt the autoinhibitory activity of the JM domain, resulting in the constitutive activation of FLT3. Also, point mutations such as Asp835 or Asn841 within the activation loop of the FLT3 kinase domain lead to constitutive activation of the receptor, resulting in FLT3 autophosphorylation and phosphorylation of downstream targets.
