Kpna2 functions in nuclear protein import as an adapter protein and binds directly to substrates containing either a simple or bipartite NLS motif. Importin-alpha is shown to exist in a monomeric autoinhibited state. Association with importin-beta increases the affinity for NLSs. The truncation of N-terminal autoinhibitory region containing an importin-beta-binding domain elevated the affinity of Kpna2 to NLS peptides.
